川贝母基源植物鳞茎内生真菌产生物碱菌株筛选及活性测定

川贝母基源植物鳞茎内生真菌产生物碱菌株筛选及活性测定
Screening of alkaloid-producing strains and activity determination of endophytic fungi from the bulbs of Fritillaria cirrhosa plants
从川贝母基源植物鳞茎内生真菌中筛选能代谢产生川贝母生物碱活性成分的菌株,并对其进行抗氧化活性和抑菌活性评价.结合生物碱显色反应、薄层层析(TLC)和高效液相-蒸发光散热(HPLC-ELSD)方法检测内生真菌菌株发酵产物的生物碱成分,采用DPPH自由基清除力测定、ABTS自由基清除力测定和铁离子还原/抗氧化能力测定(FRAP)方法对菌株发酵产物进行抗氧化活性评价,运用平板打孔法对其进行抑菌活性评价.研究发现4个镰刀菌属内生真菌菌株(Fusarium tricinctum、 Fu. oxysporum、Fu. solani和Fu. sp.)能产生与川贝母相近的西贝母碱、西贝母碱苷和贝母辛生物碱活性成分;1个镰刀菌属菌株(Fu. petersiae)的三氯甲烷提取成分具有较强的抗氧化活性,1个白腐菌属菌株(Phanerochaete sp.)的总生物碱成分具有较好的抑菌活性.综上所述,镰刀菌属的内生真菌可能产生川贝母生物碱活性成分,且其发酵产物具有较好的抗氧化活性,值得进一步研究和开发应用.
Endophytic fungal strains from the bulbs of Fritillaria cirrhosa plants were screened for their ability to metabolize and produce the alkaloid active ingredients of F. cirrhosa, and their antioxidant and antibacterial activities were evaluated. Alkaloid color reaction, thin layer chromatography ((TLC), and high - performance liquid chromatography - evaporative light scattering detection (HPLC - ELSD) were used to detect the alkaloid components in the fermentation products of the endophytic strains. The antioxidant activity of the fermentation products was evaluated by determining the DPPH free radical scavenging capacity, ABTS free radical scavenging capacity, and ferric reducing antioxidant power (FRAP), while the antibacterial activity was evaluated using the agar well diffusion method. It was found that four endophytic fungi strains of Fusarium (Fu. tricinctum,Fu. oxysporum,Fu. solani and Fu. sp.) were able to produce the alkaloid active components of sipeimine, sipeimine - 3 - β - D - glucoside, and peimisine, which are the same as or similar to those found in F. cirrhosa. The trichloromethane extract of a Fu. petersiae strain exhibited strong antioxidant activity, and the total alkaloid components of a Phanerochaete sp. strain showed good antibacterial activity. In conclusion, endophytic fungi of the genus Fusarium demonstrate potential for producing alkaloid active ingredients characteristic of F. cirrhosa, and their fermentation products exhibit notable antioxidant activity, warranting further research and development for practical applications.
